Low Attachment 3D Assay Flowchips

Low Attachment 3D Assay Flowchip consumables have been designed for experiments with 3D cell models such as organoids, spheroids and 3D tumor models to use with our Pu·MA Systems.

The strategic partnership using our proprietary microfluidics combined with ISurCell™ ULA coating from ISurTec®, enables applications such as biomarker detection by immuno-fluorescence staining or co-culture assays with immune cells. 3D cell models are formed, assayed, analyzed, and imaged all within the same flowchip.

Cell Culture Capabilities

Low Attachment 3D Assay Flowchips are to be used within the Pu·MA Systems. They are miniaturized consumables like cell culture plates but with the added feature of microfluidic channels that connect the sample well to adjacent reagent wells. This enables media and reagent exchanges without the need for pipetting.

Shown here is a schematic of how you can assemble 3D cell models within the flowchips from a cell suspension, assay the 3D cell models and then image the samples within the flowchips using confocal or fluorescence microscopy.

 

ISurCell™ ULA Coating from ISurTec®

The ISurCell ULA coating is an ultrathin, non-cytotoxic, non-auto fluorescent, xeno-free, optically clear polymer coating from ISurTec. It is highly effective at preventing both protein and cell attachment to the coated flowchips. The Low Attachment 3D Assay Flowchips have have been stably coated for cell culture conditions using the ISurCell ULA coating, but should not be autoclaved or rinsed with organic solvents such as ethanol.

Comparison Data

Here is comparison of data of culturing 3D cell models in Low Attachment 3D Assay Flowchips (within the Pu·MA System) along side a commercial 96-well ULA U-bottom cell culture plate.  HCT116 cells (3000-5000 cells) were seeded as a single cell suspense and cultured over a 72 hour period and then stained using standard Live/Dead stains. The confocal images were acquired using CellVoyager CQ1 Benchtop High-Content Analysis System (Yokogawa).

Low Attachment 3D Assay Flowchips are consumables that are to be used within the Pu·MA Systems designed using proprietary microfluidics. 4 flowchips fit in the holder and they are designed for reagent and media exchanges via microfluidic channels.
