3D Sphere Scaffold Enables Non-Compacting Organoid Establishment of Stromal Cells

Dr. Zhou is a Research Scholar (postdoc) in Dr. Anindita (Oni) Basu’s group at University of Chicago. Her current work is focussed on Crohn’s disease and the inflammatory responses in the gastro-intestinal tract. She received her PhD from University of Maryland.
Webinar Summary:
The stromal compartment coordinates inter-and-cross-system responses in human intestines, including wound healing and morphology remodeling. Importantly, the plasticity of these cells allows them to migrate and adapt acutely towards the site of stimulation or tissue damage. Unlike macrophages or lymphocytes, the stromal cells often act in a team. Thus, to better study the functions of these stroma, a micro-fluidic (water in oil co-flow) based organoid generation system was developed. It enables high throughput organoid production with Matrigel and provides flexibility of cell density and organoid sizing. Consequently, the 3D organoid is compatible with extended culture and maintains the complexity of cell identity and transcriptomes that better represents the stromal habitat in vivo.
Research findings from this development to further understand human intestinal wound healing and remodeling using 3D organoid cultures will be presented.
